What is the Metabolism and Transporter Module?
The Metabolism and Transporter Module is an optional module that extends the capabilities of
GastroPlus to include saturable metabolism (gut, liver, and/or any PBPK tissue), saturable carrier-mediated transport by both influx and efflux proteins, and metabolite tracking.
This module calculates Michaelis-Menten rates for gut and liver (or any PBPK tissue) metabolism and for carrier-mediated transport (influx or efflux) based on input values for Vmax and Km. You can provide Vmax and Km values for each enzyme/transporter independently, or you can lump them into a single effective Vmax and Km, depending on your data. The distribution factors on the Physiology tab are automatically loaded for recognized gut enzymes and transporters, and provide the relative amounts of enzymes or transporters in the various ACAT gut model compartments. The Vmax and Km scale factors on the Pharmacokinetics tab are provided to allow fitting nonlinear kinetic models to your data by scaling each of four functions: liver metabolism, gut metabolism, influx transport, and efflux transport.
The module comes with a convenient unit converter form that allows you to quickly transform your in vitro clearance measurements (intrinsic clearance (CLint), Km and Vmax, or metabolic half-life values) into in vivo values required for entry into GastroPlus.

You may now track metabolites formed in gut or any tissue in your compartmental PK or PBPK models! Simply establish records in your GastroPlus database for the parent and metabolite(s), and link them through our Enzyme Table. Account for multiple metabolites from the parent, metabolites of metabolites... the program's flexibility allows you to model almost any situation!
