Title: BIOLOGXsym – Use of Liver MPS and QST modeling to simulate biologics-inducded liver injury

Quantitative systems toxicology (QST) modeling incorporates dynamic pharmacokinetic exposure and drugs’ ability to alter key hepatocyte pathways, informed by mechanistic studies from human-derived in vitro systems, to predict the frequency and severity of drug-induced liver injury (DILI) in simulated patient populations. In vitro human microphysiology systems are 3D tissue/organ biomimetics that can further improve our mechanistic understanding about hepatotoxicity mediated by small molecules as well as biologics. In this talk, application of QST modeling and mechanistic toxicity data from Liver Acinus MicroPhysiology System (LAMPS) to investigate biologics-mediated hepatotoxicity will be presented using tocilizumab as a case study.
